This patch of American mandrake was located in the forest near Great Bear Mound in Effigy Mounds National Monument, IA.

In 1975, I observed the late Evelyn Snook (an herbologist, formerly of Lewistown, PA) use American mandrake root extracts (along with celandine and comfrey) to cure a teenage boy of Wilms' tumor; he had contracted the tumor after recovering from mononucleosis. After several days of applying poultices, the tumor in his groin disappeared.

In the early 1980s, I reported this cancer treatment to Dr. James A. Duke at the USDA-ARS Federal Research Center in Beltsville, MD, and I urged him to reinvestigate American mandrake as a potential source of podophyllotoxin-based anti-cancer drugs. After NCI chemists determined there were not enough of the active constituents in American mandrake, I urged Dr. Duke to collect samples of related Podophyllum species in other countries. Eventually, Duke's renewed research led to the discovery of Indian mandrake (Podophyllum emodi) as a source of two new anti-cancer drug compounds.

In 1989, I wrote an update about the experimental podophyllotoxin drugs, VM-26 and VP-16-216, derived from Indian mandrake: "VM-26 (etoposide) has proved effective against testicular cancer, lymphoma, small-cell lung cancer, and leukemia; VP-16-213 (teniposide) has shown evidence of clinical activity against brain tumors and non-Hodgkin's lymphoma in adults, and against neuroblastoma and acute lymphocyte leukemia in children …" (Oldfield, M.L. 1989.
